Different Approaches for Silver Nanoparticle Sterilization for Administration to Cell Culture

In recent years, there has been increased interest in the use of nanostructures various industries, such as food, textile, pharmaceutical, electronics, and chemical industries. Most these applications require a proper preparation specific nanomaterials. this study, we characterized silver nanoparticles (AgNPs) stabilized with polyvinylpyrrolidone prepared aqueous suspensions using dynamic light scattering, atomic force microscopy, transmission electron microscopy. We aimed to compare influence different AgNP procedures, specifically autoclaving, sonication, combination both, on agglomeration nanoparticles. Additionally, toxicity NPs after selected sterilization methods toward EA.hy926 endothelial cell line was determined trypan blue labeling, 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) tetrazolium salt reduction tests, 3-[4,5-dimethylthiazol-2yl]-2,5-diphenyltetrazolium bromide (MTT) test. Based obtained results, concluded that cells exposed AgNPs sonication had similar viability values above 80% across all cellular tests. Conversely, autoclaved solutions differed. From XTT observed falsely value 100% simultaneous increase XTT-measured absorbance for medium autoclaving. However, other tests showed below 25%. The results prove importance selecting an appropriate method measuring viability, especially previously sterilized
